The thioredoxin-targeted portion of the ATP synthase consists of two cysteines located on the central axis subunit γ. The molecular architecture of the chloroplast ATP synthase, which confers redox regulatory properties requires further investigation, in light of the molecular structure of the enzyme complex as well as the physiological significance of the regulation system. Three of the genes located in one cluster in cyanobacteria, however, are localized in the nuclear rather than the chloroplast genomes of plants. Regulation of the activity of the chloroplast ATP synthase is largely accomplished by the chloroplast thioredoxin system, the main redox regulation system in chloroplasts, which is directly coupled to the photosynthetic reaction. The terms FoF1-ATP synthase and FoF1-ATPase are also used. In chloroplasts, ATP synthase receives protons from thylakoid lumen, which volume is small as compared to the mitochondrial intermembrane space (IMS) and which pH value can drop to the values below 5 (Oja et al., 1999), while in the mitochondrial IMS it drops only slightly below 7 (Moore and Rich, 1985; Porcelli et al., 2005). This process occurs within the chloroplasts, on the thylakoid membranes that are stacked into structures called grana. doi: 10.1016/j.molcel.2020.09.029. J Bioenerg Biomembr. 1. Fig. Abstract The chloroplast adenosine triphosphate (ATP) synthase is located in the thylakoid membrane and synthesizes ATP from adenosine diphosphate and inorganic phosphate at the expense of the electrochemical proton gradient formed by light-dependent electron flow. ATP is mainly produced in the mitochondria and is an important enzyme that provides energy for the cell to use through the synthesis of adenosine … The bacterial enzymes are essentially the same in structure and function as those from mitochondria of animals, plants and fungi, and the chloroplasts of plants. [Figure][4] Cryo-EM structure of the chloroplast ATP synthase.  |  2019 Oct 22;116(43):21900-21906. doi: 10.1073/pnas.1913099116. In total, 85% of the ATP synthases are monomers and the remainder form random assemblies of two or more copies. In chloroplasts, ATP synthase genes are present in two gene clusters, atpI/H/F/A and atpB/E, both of which are processed into several different transcripts (Supplemental Fig. One may also ask, can chloroplast make ATP? Commun Biol. The cyanobacterial ATP synthase Chemiosmosis - The Mechanism of ATP Synthesis in Chloroplasts The thylakoid membrane is composed of a phospholipid bilayer (color phospholipids "B" light blue) and photosystem I and photosystem II. The resulting acidification of the lumen regulates both light harvesting, via the qE mechanism, and photosynthetic electron transfer through the cytochrome b6f complex. COVID-19 is an emerging, rapidly evolving situation. Chloroplasts contain chlorophyll, two types to be exact: a and b. Structure of mycobacterial ATP synthase bound to the tuberculosis drug bedaquiline. 2016 Sep;87(6):654-63. doi: 10.1111/tpj.13226. The orientation is quite easy to remember, for the bacterium need ATP to be synthesized inside the cell, not outside. Recall where ATP synthase and the Electron Transport chain are found in both chloroplasts and mitochondria. Bacteria and archaea, which make up the prokaryotes, do not have mitochondria but produce ATP through similar cellular respiration processes in their plasma membrane. Fig. The size of granal thylakoids was determined for Arabidopsis as 4 nm (stacking repeat distance) to 5 nm (diameter) in darkness, increasing to 19 nm in width and to 9–10 … See this image and copyright information in PMC. The features of ATP synthase nanomachines are remarkably similar in chloroplasts and mitochondria, considering their evolutionary distance of a billion years or more. Hisabori T, Ueoka-Nakanishi H, Konno H, Koyama F. FEBS Lett. In light … This operon is transcribed by the plastid-encoded RNA polymerase starting from two promoters, which are upstream and within the operon, respectively, and harbors four potential sites for RNA-binding proteins. Epub 2020 Aug 31. 3. The dynamic environment of ATP synthase in chloroplasts is established in a different (and in most aspects opposite) way as compared to mitochondria. The structure, activities, and mechanism of the chloroplast ATP synthase are discussed. The early ancestory of the enzyme is seen in the fact that the Archaea have an enzyme which is clearly closely related, but has … Copyright © 2018 The Authors, some rights reserved; exclusive licensee American Association for the Advancement of Science. namely, within whole isolated chloroplasts, the ATP synthase is rapidly deactivated in the dark, although broken chloroplasts (thylakoid particles) remain active for several hours after wash-ing. Enzyme that synthesizes ATP. One possible explanation for the reduced ATP synthase level in bfa3 is that the maturation or translation initiation of these genes is disrupted. The chloroplast adenosine triphosphate (ATP) synthase uses the electrochemical proton gradient generated by photosynthesis to produce ATP, the energy currency of all cells. HHS Richter ML, Samra HS, He F, Giessel AJ, Kuczera KK. We found that stroma and grana thylakoids are connected at the grana margins by staggered lamellar membrane protrusions. Inabe K, Kondo K, Yoshida K, Wakabayashi KI, Hisabori T. J Biol Chem. The activity of the molecular motor enzyme, chloroplast ATP synthase, is regulated in a redox-dependent manner.
